The six analyses complement point-of-care testing on the Igloo reader with what it does not show: individual predisposition and regulatory processes that change over time.

Two layers: genetics and epigenetics

Genetic variants describe lifelong, unchangeable predisposition. Epigenetic markers — microRNA patterns and DNA methylation — describe the current regulatory state and respond to nutrition, exercise, sleep and strain. That is precisely why they can be measured over time.

Sample material: dried blood card

A few drops of capillary blood from the fingertip are applied to a dried blood card. No venipuncture, no centrifuge, no cold chain — sampling is possible in the practice or at home.

Laboratory and reference data

Analysis is carried out by our cooperation partner, a specialised European laboratory with more than 15 years of research in this field. Results are placed against more than 60,000 validated reference values. Quality management to ISO 9001:2015.

What is analysed

The analysis evaluates which distribution of macronutrients suits the individual predisposition, supported by specific portion and recipe suggestions. Because genetic variants do not explain weight behaviour on their own, epigenetic markers of DNA stability and inflammation are assessed in addition: only those respond to lifestyle and work as a measure of success.

What is analysed

Biological age clock

An exploratory lifestyle signal from three microRNAs — explicitly not an age determination.

What is analysed

Three microRNAs are determined by quantitative PCR, normalised against miR-93 and combined into a single value: the percentage deviation of biological from chronological age. The underlying model is published but declared a pilot study — the output is an exploratory signal, not a robust biological age.

What is analysed

Telomeres shorten with each cell division and are regarded as a marker of cellular ageing. Measurement is performed from dried capillary blood, reported in kilobase pairs and placed against an age- and sex-matched reference population. qPCR values vary considerably between runs and laboratories — the trend carries the message, not the single value.

Evidence base

What is established — and what is not

This field is heavily over-promised. Anyone presenting these analyses should know the limits — otherwise they collapse at the first critical question. With professionals, candour is an advantage, not a risk.

Well established

  • Lifestyle changes epigenetic markers

    Randomised pilot trial over 8 weeks: epigenetic age on average 3.2 years lower than in the control group (Fitzgerald et al., Aging 2021). This measured DNA methylation, not microRNA.

  • Inflammation as an ageing axis

    "Inflammaging" is an established concept; miR-21 is described as a circulating inflammation marker (Olivieri et al. 2012).

  • microRNAs are stable in dried blood

    microRNAs are considered more robust than mRNA and remain comparatively stable in dried samples — the sampling route itself is sound.

  • The established standard for biological age

    That is methylation clocks: Horvath, PhenoAge, GrimAge and DunedinPACE. DunedinPACE shows high test-retest reliability and associations with morbidity and mortality.

Preliminary

  • microRNA age clock

    The published model (n = 539) explains about 5 % of the variance in age with a mean absolute error of 10.4 years; two of the three markers did not correlate significantly with chronological age. As an age determination the value does not hold.

  • Telomere length by qPCR

    The method varies considerably between runs and laboratories; DNA extraction, storage and cell composition systematically affect the result. Assay variation from dried blood is higher than from venous blood.

  • Gene-based dietary advice

    The Food4Me study found no difference in weight loss between levels of personalisation including genetics. The benefit lies in structure, conversation and adherence — not in superiority over good standard counselling.

  • microRNA as stress markers

    There is work on individual microRNAs and acute as well as chronic stress, but no established cut-offs and no diagnostic validation.

What professional users ask

What is the difference between genetics and epigenetics?
Genetic variants are unchangeable for life and describe predisposition — they are recorded once. Epigenetic markers describe the current regulatory state, respond to nutrition, exercise, sleep and strain, and can therefore be measured over time. Only the epigenetic layer works as a measure of success.
How is the sample taken?
A few drops of capillary blood from the fingertip go onto a dried blood card and dry there. No venipuncture, no centrifuge, no cold chain — sampling is possible in the practice or at home. The card is sent to the partner laboratory in the prepared envelope.
When does a follow-up measurement make sense?
After 4 to 6 months at the earliest and under conditions as similar as possible. The trend carries the message, not the single value — an initial and a follow-up measurement together form a plannable programme. Acute infections shift inflammation markers, so the timing should be chosen deliberately.
Do these analyses replace clinical diagnostics?
No. They are consultation and monitoring tools, not a medical diagnosis and not a substitute for clinical assessment. Where findings suggest a condition requiring treatment, the person belongs in medical or psychotherapeutic assessment.
What are the legal requirements?
In Germany, genetic analyses are subject to the Genetic Diagnostics Act: the person examined must be informed and give written consent before sampling, and this must be documented. It applies to Metabolic Health and to the Plus variant of Healthy Sport.
How robust are the results?
That differs by analysis, and we state it openly. The sampling route itself is sound, as is the monitoring approach. The microRNA age clock, by contrast, is exploratory, and qPCR telomere values vary considerably. The evidence sheet separates established from preliminary evidence and names the sources.
What do the analyses cost?
B2B terms on request, tiered by volume and contract model. There is no public consumer price list. Test cards, shipping material and report delivery are part of the offer.
